General anesthetic (GA) spans a wide range of chemical structures, such as monatomic molecules, barbital acids, phenols, ethers, and alkanes. GA has a weak binding affinity, and the affinity has a characteristic that correlates with the solubility in olive oil rather than the molecular shape. The GA binding site of GABAAR is common to GAs and exists in the transmembrane domain of the GABAAR intersubunit. In this study, the mechanism of GA binding, which allows binding of various GAs with intersubunit selectivity, was elucidated from the hydration analysis of the binding site. Regardless of the diverse GA chemical structures, a strong correlation was observed between the binding free energy and total dehydration number of the binding process. The GA binding free energy was more involved in the binding dehydration and showed molecular recognition that allowed for the binding of various GA structures via binding site hydration. We regarded the GA substitution for the interfacial water molecule of the binding site as a dissolution into the interfacial hydration layer. The elucidation of the GA binding mechanism mediated by hydration at the GABAAR common binding site provides a rationale for the combined use of anesthetics in medical practice and its combination adjustments via drug interactions.","subitem_description_type":"Abstract"}]},"item_4_description_42":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"subitem_description":"Journal Article","subitem_description_type":"Other"}]},"item_4_publisher_32":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"日本生物物理学会"}]},"item_4_relation_11":{"attribute_name":"DOI","attribute_value_mlt":[{"subitem_relation_name":[{"subitem_relation_name_text":"10.2142/biophysico.bppb-v20.0005"}],"subitem_relation_type":"isIdenticalTo","subitem_relation_type_id":{"subitem_relation_type_id_text":"https://doi.org/10.2142/biophysico.bppb-v20.0005","subitem_relation_type_select":"DOI"}}]},"item_4_rights_12":{"attribute_name":"権利","attribute_value_mlt":[{"subitem_rights":"© 2023 THE BIOPHYSICAL SOCIETY OF JAPAN"}]},"item_4_source_id_7":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"2189-4779","subitem_source_identifier_type":"ISSN"}]},"item_4_version_type_15":{"attribute_name":"著者版フラグ","attribute_value_mlt":[{"subitem_version_resource":"http://purl.org/coar/version/c_970fb48d4fbd8a85","subitem_version_type":"VoR"}]},"item_creator":{"attribute_name":"著者","attribute_type":"creator","attribute_value_mlt":[{"creatorNames":[{"creatorName":"SETO, Tomoyoshi"}],"nameIdentifiers":[{},{},{}]}]},"item_files":{"attribute_name":"ファイル情報","attribute_type":"file","attribute_value_mlt":[{"accessrole":"open_date","date":[{"dateType":"Available","dateValue":"2023-06-05"}],"displaytype":"detail","filename":"biophysico.bppb-v20.0005.pdf","filesize":[{"value":"2.4 MB"}],"format":"application/pdf","license_note":"This article is licensed under the Creative Commons Attribution-NonCommercial-ShareAlike 4.0 International License.\nTo view a copy of this license, visit https://creativecommons.org/licenses/by-nc-sa/4.0/.","licensetype":"license_note","mimetype":"application/pdf","url":{"label":"biophysico.bppb-v20.0005","url":"https://shiga-med.repo.nii.ac.jp/record/4499/files/biophysico.bppb-v20.0005.pdf"},"version_id":"e5a31d6b-0edb-41b7-8302-129b9df37e25"}]},"item_keyword":{"attribute_name":"キーワード","attribute_value_mlt":[{"subitem_subject":"GABAA","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"receptor","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"general anesthetics","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"molecular recognition","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"protein hydration","subitem_subject_language":"en","subitem_subject_scheme":"Other"},{"subitem_subject":"dissolution","subitem_subject_language":"en","subitem_subject_scheme":"Other"}]},"item_language":{"attribute_name":"言語","attribute_value_mlt":[{"subitem_language":"eng"}]},"item_resource_type":{"attribute_name":"資源タイプ","attribute_value_mlt":[{"resourcetype":"journal article","resourceuri":"http://purl.org/coar/resource_type/c_6501"}]},"item_title":"General anesthetic binding mode via hydration with weak affinity and molecular discrimination: General anesthetic dissolution in interfacial water of the common binding site of GABAA receptor","item_titles":{"attribute_name":"タイトル","attribute_value_mlt":[{"subitem_title":"General anesthetic binding mode via hydration with weak affinity and molecular discrimination: General anesthetic dissolution in interfacial water of the common binding site of GABAA receptor"}]},"item_type_id":"4","owner":"11","path":["254"],"pubdate":{"attribute_name":"公開日","attribute_value":"2023-06-05"},"publish_date":"2023-06-05","publish_status":"0","recid":"4499","relation_version_is_last":true,"title":["General anesthetic binding mode via hydration with weak affinity and molecular discrimination: General anesthetic dissolution in interfacial water of the common binding site of GABAA receptor"],"weko_creator_id":"11","weko_shared_id":-1},"updated":"2023-06-19T11:06:18.276438+00:00"}
